Microeukaryote community patterns along an [O.sub.2]/[H.sub.2]S gradient in a supersulfidic anoxic Fjord (Framvaren, Norway)
Article Abstract:
Cultivation-independent 18S rRNA survey is conducted in the superanoxic Framvaren Fjord in Norway and three clone libraries are generated along the steep [O.sub.2]/[H.sub.2]S gradient by using the multiple-primer approach in order to find the fine-scale architecture of anoxic protistan communities. The statistical and phylogenetic analyses of the three libraries have revealed the fine-scale architecture of anoxic protisan communities, which might display adaptation to different environmental conditions along the [O.sub.2]/[H.sub.2]S gradient.

Cellular identification of a novel uncultured marine stramenopile (MAST-12 Clade) small-subunit rRNA gene sequence from a Norwegian estuary by use of fluorescence in situ hybridization-scanning electron microscopy
Article Abstract:
Fluorescence in situ hybridization is used with an oxygen-depleted incubation culture obtained from an estuarine microbial mat in order to determine the cellular identity and lifestyle of a novel phylogenetic lineage branching within the uncultured marine stramenopiles (MAST-12) clade. The morphological characteristics have helped in isolating a representative of a novel stramenopile group in order to study its ultrastructure, physiological capadbilities and ecological role in the environment.

Methodology of protistian discovery: From rRNA detection to quality scanning electron microscope images
Article Abstract:
A new combination of the modified 18S rRNA, fluorescence in situ hybridization (FISH), and scanning electron microscopy methodologies is described. The combination allows identification of target cells by FISH to be followed by fine-quality SEM studies, with both performed on the same microscopic preparations.
